圆二色谱(Circular Dichroism, CD)技术是一种用于分析手性分子的二级结构、构象变化及其热稳定性的光谱学方法,广泛应用于蛋白质、核酸等生物大分子的研究中。该技术基于手性分子对左旋和右旋圆偏振光吸收差异的原理,不同的二级结构(如α-螺旋、β-折叠和无规则卷曲)在远紫外区(约190–250 nm)具有特征性的CD谱图。CD实验一般在溶液状态下进行,样品无需标记或结晶,适用于原位检测。通过测量不同波长下的圆二色信号,可快速获取样品的二级结构组成,并可进一步结合热熔解实验(温度梯度CD扫描)评估其热稳定性(如Tm值)。此外,CD还常用于比较突变体与野生型蛋白的构象差异、监测复合物形成及蛋白质折叠过程。CD数据通常可用于定性判断,也可借助拟合软件进行二级结构含量的定量估算。由于操作简便、样品用量少、数据获取快速,CD技术已成为生物大分子结构功能研究中的常规手段之一。

Dear all,
Circular Dichroism (CD) spectroscopy is a widely used spectroscopic technique for analyzing the secondary structure, conformational changes, and thermal stability of chiral biomolecules such as proteins and nucleic acids. This technique is based on the differential absorption of left- and right-handed circularly polarized light by chiral molecules. Distinct secondary structures, includingα-helices,β-sheets, and random coils, exhibit characteristic CD spectra in the far-UV region (approximately 190–250 nm).CD measurements are typically performed in solution, without the need for labeling or crystallization, making the technique suitable for in situ structural analysis. By recording the CD signals across different wavelengths, researchers can rapidly assess the secondary structure composition of a sample. Furthermore, thermal denaturation experiments (e.g., temperature-dependent CD scans) can be used to evaluate the thermal stability of biomolecules, such as determining the melting temperature (Tm).In addition to structural characterization, CD is commonly employed to compare the conformational differences between wild-type and mutant proteins, monitor complex formation, and study protein folding processes. CD data can be interpreted qualitatively or analyzed quantitatively using software-based secondary structure deconvolution methods. Due to its simplicity, low sample requirements, and rapid data acquisition, CD spectroscopy has become a routine and essential tool in the structural and functional analysis of biomacromolecules.
